1-Determine the number of interaction sites (stoichiometry), constant Stern-Volmer and the binding constant by suppression tests fluorescence of genistein (GEN), doxorubicin (DOX) and Human Serum Albumin (HSA). Triplicate at 25°C and triplicate at 37°C
2-Carry out competition assays between DOX and GEN ligands by HSA through fluorescence quenching assays. Triplicate at 25°C and triplicate at 37°C
3-Characterize the stabilities of potential HSA-DOX, HSA GEN and GEN-HSA-DOX complexes by fluorescence spectroscopy. Triplicate at 25°C and triplicate at 37°C
Materials:
Doxorubicin D1515 Sigma Aldrich (use Tris buffer, not PBS buffer. PBS precipitate doxorubicin)
Genistein G6649 Sigma Aldrich
HSA 99% pure A3782 Sigma Aldrich
